should TDS be deducted on job charges even if no contract exist? also if one bill amount is rs. 45000? and all bill amount totals to rs 250000?

In this case the provision of tax deduction will be applicable. The work done must be in nature of work contract having a formal agreement or not is irrelevant for applicability of tax deduction.
