Ram sells a property to Shyam for `10,00,000 which is subject to mortgage to Mohan for `20,00,000 and unpaid interest of `4,00,000. Stamp duty is payable on `34,00,000 or on 100000?

What you want is not clear. Anyhow, in case of sale there will be separate registration in favour of Shyam and stamp duty is to be paid on 10L.
After the sale shyam becomes the owner then he has to execute a deed of mortgae in favor of Mohan and separate stamp duty is payable on 24L
