One of our client has been registered under Registrar of Society, Uttar Pradesh while Income Tax Return has been filed in Delhi as per Delhi address and want to apply application for 12A and 80G Exemption in Delhi. Is it possible.

These applications are filed where Income Tax jurisdictions lies. So may file in Delhi, if you have already filed returns in Delhi jurisdiction.
