If any Director resigns during the year i.e. before completion of year, is he required to submit his disclosure of interet through Form-24AA before BOD?

Yes, if he resigns in the month of Feb or March he needs to make a disclosure in Form 24AA before BOD and According to Section 299 of the Companies Act, 1956, the Directors of the Company place Form 24AA, when they are appointed in the Board and every year at the last month of the Financial Year after its renewal.
