Suppose a proprietor, who is liable for audit u/s 44AB, gives Rs. 50,000 to brother as interest and no TDS was deducted. And the brother’s income is less than Rs.250,000.Can his brother submit form 15g to the proprietor?

first off all Form 15G/H and Form 13 must have assent from the Assessing officer. It's not like that you your self prepare these forms and submit to the payer. Alternatively if your brother's ITR has been filed before due date then you can take certificate from CA [Form 26A]
