How could i calculate the turnover from speculative income, as sheet which i got for trading during the year only contains the net profit from speculative income?

You can obtain it from your broker as mostly every broker gives statement which contains date purchase price selling price and number of shares so you can calculate it with that.
Should i sumup the number sale value, as i haven't got, need that for itr-3, there is loss - 4000?
Yes for finding the turnover you have to add the sale value of all the transactions
