I would like to know whether enclosing of any proof is required at all since returns are to be filed without any enclosures.

It is enough if you provide doctor's certificate along with Form 10IA. The certificate should be from doctors of prescribed medical authority. The claim of either Rs.50K or 75K is a flat deduction and irrespective of amount of expenditure that has been incurred on handicapped dependant. No proof of expenditure need to be attached to support deduction claim.
