I wanted to know that if buyer is purchasing the Residential house directly from builder, whether buyer is required to deduct TDS from payment to builder.

Yeah, buyer has to be deduct TDS if consideration is >= 50 lacs
Yes as sec 194IA -Transfer of certain immovable property other than agriculture land(w.e.f 1-6-2013) which exceeds 50laks then TDS @ 1% be deducted.
