Yes, as per instructions attached to form REG-01 i.e. Application for New registration has to be digitally signed as under :-
Application filed by undermentioned persons shall be signed digitally:-
Sr. No Type of Applicant
1. Private Limited Company
2. Public Limited Company
3. Public Sector Undertaking
4. Unlimited Company
5. Limited Liability Partnership Foreign Company
6. Foreign Limited Liability Partnership
Persons Other than above should either sign the application Digitally or as may be notified
It also specifies as to who should sign the application :-

There is also an option to E-sign if you have aadhar card.

Any body knows when we can submit with E-Signature as it is still block.

The E-signature oprtion is now available. Please have a check.
However, as per the GST helpline (1800 103 9271-Toll Free) it will carry a period charge, the details of which is not available at the moment.
So, request to look into the same carefully and sign

DSC or Digital Signature is a physical signature in an electronic format.Digital Signatures are used in India for online transactions such as Filing Annual Return, Company or LLP Incorporation, Income Tax E-Filing, E-Tenders, etc. There are 3 sorts of Digital Signatures, Class I, Class II and Class III Digital Signatures. Class I Digital Signature is utilized for securing email communications. Class II digital signature is utilized for company registrations, IT Return E-filing, Obtaining DIN, DPIN etc. Class III digital signature registration are used for E-tendering and participating in E-Auctions.
Now you can create digital Signature certificate (DSC) class II just rupees 500/- only.
