when does the same reflect in 2a? And what about invoice amendement, when does the same reflect in 2A?

when you filed the return
you mean after the other party files gstr 1?
YES
actually when he enters the invoice it will be shown with counter party status as NO
but it will be YES after filing of return by counter party
