if any company providing the job to a professional on the package basis....is package cost include gratuity???

Yes it will include...recently all companies which is following GTA 1972 are including gratuity cost in CTC.

BUT actual gratuity applicable after five year of complition of employee than how they determine that the employee will work for more than five year?????
