Dose the additional director need to give a fresh consent letter to act as a director in case of regularisation of his appointment as director at the AGM. This is for the public company

No, consent need not be given again.
The consent given by the Additional Director is not till the date of AGM. The consent letter only specifies that the respective person is interested and is agreeing to become a director in the company.

Agree with Mitali.
However, I would like to share further some info which I read in 'Mr. Jayaraman's book' - if additional director appointed as a regular director in forthcoming AGM, he will have to comply with Sec. 257 i.e. notice must be given to Company that he is interested in directorship of Company.

If DIR-2 is 06.11.20 and appointment was made on 06.02.2021 can we take that DIR-2 as his consent to act as director or do we need to take new DIR-2 ? Means is there any validity period for DIR-2 ?
