The query asked is whether ITR - V acknowledgement can be used for taking acknowledgement in respect of returns filed in the income tax ward office and not efiled.

No, we cannot use ITR-V as acknowledgement receipt for paper return. IT dept is yet to notify the format for acknowledgement of paper return.
ITR-V is to be used only when the return of Income is filed electronically & without digital signature.

ITR - V acknowledgement for return efiled without digital signature only. For rest i. e. return filing in ward office like last year acknowledgement only to be notified.
